RESUMO

One of the important factors influencing the microbial community of ruminants, besides environment or diet, are breed and physiology. Therefore, the purpose of this study was to assess these changes in the levels of basic microbial phyla and families. For this study, qPCR analysis was performed to determine the level of bacteria (Firmicutes, Bacteroidetes, Actinobacteria, Proteobacteria clusters and Clostridiaceae, Lactobacillaceae families) in the feces of ewes of three native Polish sheep breeds (Polish Lowland Sheep (PON), Swiniarka Sheep (SW), and synthetic line BCP) at different physiological periods (conception, early pregnancy, lambing, end of lactation). The animals were kept in the same environment and were at the same age (2-years). The results showed a significant effect of both breed (p = 0.038) and physiological period (p < 0.05, p < 0.01) on the levels of bacteria analyzed. The breed showed differences across physiological periods. The influence of the race factor was noted primarily between the BCP synthetic line and the other two breeds (differences in terms of all analyzed clusters and families except Actinobacteria phyla). In the case of SW and PON, however, the observed differences were only at the level of Proteobacteria cluster and Clostridiaceae family. On the other hand, the early pregnant and lambing periods were the most microbiologically diverse in terms of the analyzed clusters and families of bacteria.

A well-functioning immune system is the basis for protection against infectious and metabolic diseases, and a smooth return to homeostasis. The periparturient period is considered critical because major changes in the endocrine, behavioural, digestive, and immune systems dysregulate immune function, leading to immunosuppression. With age, the immune system could become dysregulated. The purpose of the present investigation was to compare changes in plasma concentrations of acute phase proteins, cortisol, and haematological parameters in the peripheral blood of two age-related groups of healthy ewes to get a better understanding of changes around lambing. Two groups of ewes were enrolled in the study: 3-year-old (young; n = 9) and 7-year-old ewes (old; n = 9). All females were synchronised and inseminated. In blood plasma, serum amyloid A (SAA) and cortisol concentrations were measured using ELISA tests, a spectrophotometric method to determine haptoglobin (Hp), and a thrombin clottable estimation to determine the fibrinogen (Fb) concentration. The blood parameters were examined using an automated haematological analyser. In clinically healthy ewes, no significant effect of age was observed in SAA, Hp, Fb and cortisol concentration in most of analysed terms. SAA, Hp, Fb, and cortisol fluctuations typical for the periparturient period were observed. There were no age-associated differences in red or white blood cell parameters.

Many subclinical diseases associated with inflammation occur in sheep during the periparturient period. Clinical symptoms are usually detected at an advanced stage of the disease; therefore, there are considerable risks of permanent health disorders in fetuses or dams. Determination of acute phase proteins (APPs) as markers of inflammation may allow for an earlier diagnosis and effective treatment. Furthermore, multi-fetus pregnancies are often associated with hematological disturbances. The study objective was to compare plasma concentrations of serum amyloid A (SAA), haptoglobin (Hp), fibrinogen (Fb), and cortisol in ewes bearing one and two fetuses in the period from 2 weeks before to 2 weeks after parturition as well as to determine hematological parameters in peripheral blood. There was an important effect of fetal number on APP and cortisol concentrations in periparturient ewes. There was a greater concentration of SAA, Hp, Fb, and cortisol in ewes bearing two fetuses compared with those bearing one fetus. Profiles for APP and cortisol concentrations and hematological parameters were similar for ewes bearing one and two fetuses, and trends were within normal reference ranges for the periparturient period. Furthermore, there were no differences in values for hematological variables between ewes bearing one and two fetuses. with there being no ewes with anemia. In summary, separate determination of the previously undefined physiological ranges of APPs and cortisol for ewes bearing one and two fetuses may facilitate diagnosis of subclinical disorders and enable comparison of laboratory test results with different reference values for ewes bearing different numbers of fetuses.

Numerous subclinical diseases in sheep occur in the periparturient period and involve inflammatory processes; therefore, determining markers, such as acute-phase proteins (APPs), can allow an early diagnosis. Therefore, the objective of the present study was to assess changes in the plasma concentration of APPs and cortisol in clinically healthy ewes in the periparturient period for use in future studies. At the same time, haematological parameters were monitored. We showed that plasma APPs and cortisol concentrations were significantly higher in pregnant ewes than before insemination. A gradual increase in the SAA concentration was observed from the 14th day before to the day of parturition, while Hp was reduced from 2 weeks before up to 2 weeks after delivery. A significant increase in the Fb concentration was detected from the 14th day before to the 1st week after delivery. The cortisol concentration did not undergo significant changes in the periparturient period. We found an increase in the SAA and Fb concentrations and decrease in Hp in the periparturient period. The direction of the change in APPs of healthy ewes in the current study may be related to their distinct regulatory mechanisms during pregnancy. The APPs are usually altered during infection, inflammation, neoplasia, stress and trauma; therefore, knowing their reference values could help lead to an early diagnosis of subclinical forms of some diseases and pregnancy complications in ewes. The haematological analysis showed that ewes in late pregnancy and postpartum compared to dry period were under metabolic stress related to pregnancy and lactation.

The research objective was to assess the behavior patterns of the Polish Konik horse breed and the Uhruska variety of the Polish Lowland Sheep breed under a mixed-grazing system, and their relationship with climatic factors. The observation included 4 adult horses, 27 ewes with lambs and 10 primiparous ewes. The behavior of the animals and the weather conditions were recorded at 60-min intervals. Horses and sheep displayed similarities in both species-specific behavioral patterns and timing of grazing activity, and the duration of foraging sessions was mostly influenced by time of day, lower temperature and relative humidity. In addition, the two species showed more interest in the watering place in the afternoon, and drinking frequency was mainly dependent on air temperature. The animals that pastured together gradually began to mix, but at first, they remained within their species group. Both animal species can be used for environmental protection and landscape conservation making rational use of pastures within protected areas.
